On the right hand side of the bike just behind the engine, there is an oblong chrome cover on the side of the transmission. In that is a plug that takes a large allen wrench or hex key to remove the plug, 7/16". Remove the plug and you'll find a dipstick. The transmission holds 20-24 ounces of oil. Make sure the oil is up to the mark when the bike is standing straight up.

I think that this may have been changed after it left the factory. Was this bike new to you or was there a previous owner?
I am wondering if someone did this so they would know what plug was engine oil and what was transmission oil. I know that pulling the wrong plug is a hassle, and depending on if you drop it or not you may be refilling the transmission or engine oils when you didn't want to.

How to drain the oil on a 2001 harley softail heritage, and where exactly is the drain plug and how much oil should you replace it with?

There is a large hose that runs down behind the transmission on down to the very bottom of the frame behind the exhaust pipes on the right side of the frame. It usually ends down on top of the lower frame tubes with a large metal plug in the end of the hose. I think later models have the hose run to a crossmember of the frame with a plug that screws into the crossmember from underneath. Remove the plug and drain the oil. Refill with three quarts of oil with a filter change. SAE 20W50.

How do i drain oil from an '01 heritage softail?

There is a hose or steel line that comes from the bottom of the right side of the oil tank and runs down the frame. Look for a plug held in by a clamp or a screw in plug just behind the transmission on the frame. Trace it up to the bottom of the tank or vice versa.

follow the rubber tube comeing out of the bottom of oil tank,at the end is the plug..down by the swingarm
