My 2000 chevy silverado misfires upon attempting to accelerate from a stop sign or from parking. It sometimes actually sometimes sounds like it backfires and spark knocks.If push the accelerator halfway down it will fall on it's face and finally start to accelerate like it is finally getting fuel or air.I've already replaced the fuel filter and pump with no change. What steps do I take next?

It sounds like it could be an engine misfire. As the engine spins faster and faster that misfire makes itself less apparent. However on stops and low idle speeds it will be very obvious. If the check engine light is on that would also help you narrow down the issue.

Have the PCM diagnosed, it may be defective. If the engine is really misfiring and you are dead sure of that the check engine light should be blinking at you. Are you sure the light works? Does the CEL prove out when key is turned to run?
Intake manifold gasket or cracked intake manifold will cause a variety of problems including rough idle and random misfire. You can try spraying a little carb cleaner at various points around the intake manifold while the engine is idling and see if the RPMs pick up at any point. This would indicate that it is drawing air in someplace where it shouldn't. The slow acceleration is probably because the computer has set itself all out of wack and is trying to compensate, that's why you get codes like O2 sensor when it really isn't bad, it's just raw gas getting to it.
The noise is caused by the axle button being worn out, this lets the axle move back and forth and clunk, this is a unique problem with C clip retained axles, it is a noise issue but will not fail and cause any more damage, in order to fix it you must have the axles pulled and the diff carrier checked for excess wear where the axle retainning C clip rides, if the carrier is ok then replace the axles.
