Yes, I would lean toward a stuck drivers door latch. Your door open switch is located in the latch itself and the ding is a reminder your key is in the ignition so you don't lock them in the vehicle when you lock the door. even though your door is shut, the truck still thinks it is open. sometimes spraying the latch with some WD-40 will get it freed up and working again, but a new latch might be in your future.
With key on door open, hold switch in down position and rap door panel with rubber mallet...if it works, need motor, if not, check switch for power out to motor
Inspect or replace the window lock and unlock switch on the drivers door power window switch. There is a lock and unlock switch that will disable the switches to the other power window in the Jimmy. Check to make sure that switch is in the unlock position to have the other power window switch to work.

The vehicle will lock and unlock the doors automatically. 10.Hold down the LOCK and UNLOCK buttons on the transmitter simultaneously until the door locks and unlocks. This step may take up to 30 seconds. 11.Repeat step 10 NOW for each additional transmitter (including any existing transmitters). 12.Turn the ignition switch to the ON position in order to exit the transmitter programming mode.
