That's easy just disconnect the negative(-) battery teminal. Do not pry the terminal off you'll damage the battery, instead either twist the terminal until it comes of or pick up a battery post puller. I believe you can rent one at Autozone on there rent for free program.
If you have any other problems your check engine light may come back on almost instantly. If that happens you'll have to fix those problems before the light will stay off when it's reset.

Just put a used engine in and the check engine light is flashing and car is running rough.

Auto part stores will run a diagnostic for free as long as you can get your car there, it not, they may rent one. After replacing bad sensors, see where you're at, a tune up with plugs, wires, cap, rotor, air and fuel filters if needed, check them out first. Start with the sensors and go from there. Make sure your ground connections are solid with no corrosion, frayed wires or loose connections.

2000 buick lesabre runs rough, black carbon on one coil post when removing plug wire.

The coil may be bad. Was the plug wire inserted all the way in? Could be a bad coil, but also check that wire and its spark plug for any sign of damage. Not likely, but check.

Check for a cracked plug or bad wire. Make sure the plug wire isn't grounding out or leaking around the plug. Easiest way is in the dark with the engine running.

99 Buick Lesabre Check engine light flashing.I changed wires and plugs but still misfiring on one cylinder, but all plugs are firing. Have 84,000 miles and has been running great until light came on and...

From the symptoms of the problem and the precautions you have done i think its time to check the EGR valve.Clean your EGR valve and then put it again. How handy you are in ding this?

As the check engine light is on did you scan for error code which will help you more specific to locate the problem.

Fuel injectors go bad on these engines, you can find re manned ones for 30 bucks. If you have an engine light on get that read. Change your coil/wires/plugs also. Is the lower intake manifold gasket leaking coolant yet? You may want to get that replaced also. Check the TPS too (throttle positioning sensor) those will also cause your engine to act up

Engine light blinking

You may have a bad spark plug wire or plug, if that doesnt solve the problem it could be the ignition coil.
