The remote likely needs to be reprogramed. If you have both remotes you will need to take these to the dealer and have them reprogramed.
You put one key in the ignition turn it on don't start it put another key in the door and lock the door within 5 seconds hit the lock or unlock, I forget possibly twice and it should be programmed.
Put your good key in the ignition and turn to "ON", do not start the engine. Make sure all the remotes you want to use are present because they have to be programmed at the same time.
- Close the door and insert key #2 into the driver's side lock. Lock the door.
-Within 5 seconds of locking the door, press unlock 2 times on the new remote, key #2.
-(if this is remote #3, press unlock 3 times. if this is remote #4, press unlock 4 times).
-If there's a chirp or beep do not press it again. This means that the car has recognized the new keyless entry remote.
- Repeat with other remote or unlock the car and turn off - done.
SOURCE: key wont' remotely lock/unlock (new batteries)

SOURCE: Key wont open door on Chevy Colorado, only remote
Try spraying some wd40 into the lock loads of it and keep turning the key left to right might free off.
Or it could be a rod inside the door has come off so the lock wont move to unlock, the central locking has a motor to open the door that is not attached to the rod so it can work without the lock.
Take the door card/trim off and have a look at the rear of the lock, should have 1 rod for the handle, 1 for the lock mechanism and another for the inner handle. some are cables but most are rods
It might just have came off the back of the lock, they use very small plastic clips to hold the rod in place, when it gets cold the crack and fall off so the rod fall's out.
Hope this helps
