Lift the trunk up and then may have to remove some of the panel behind the light case. once you've done this, you'll have another panel/circuit board on the way, this should be easily removed by pinching the clips on the circuit panel and then the circuit panel falling loose.
locate your brake light bulb and then remove the bulb like a house bulb...push and twist.
most of the time, the owners service manual gives brief directions of how to go about and removing bulbs

Identify which side of the light bulb is out.

I like to check it in pair and save the know good bulb for trouble shooting.

You will need to empty out most part of the tank to the to the bulb compartment

You can use two know of bulb in your Audi A6.

7528LL
(LL = long life)

or the standard

7528

I do recommend to purchase a small pack of bulb grease to prevent the bulb for getting stuck inside the socket.

PLEASE WEAR protective gloves when you push and twist the bulb out of the socket.

Test the bulb before you close compartment.

Re-check for driving light and brake operation.

Good luck

I have a 1999 Audi A6 quattro my right brake light went out I went to to switch the bulb and the bulb was OK but wont come on now I can't put my shift gear into gear without turning my lights on

There's should be a shift interlock that prevents it from being put into gear unless the brakes are on; check if the fuse it good for the brake lights; it seems to somehow be tied in with the parking lights, which may be feeding into the brake light circuit.

2005 audi A6 intermittent tail light failure

I can't solve this problem, but have had the entire left tail light unit on a 2005 A6 replaced under warranty 5 times in 3 years. Once it was out of warranty, the dealer kept replacing bulbs and charging me $30 each, but it never solved the problem, even for a day (clearly looking to duck their lifetime guarantee on work they did).

Audi A6 Quattro - Looses lights

They usually have igniters that will start to give out on the light bulbs. But if they did not find any faults then it would require a long diagnosis time.

Audi Quattro A6 will not go into drive

It's the brake pedal switch. There is a switch down on the pedal arm that closes when you press the pedal down, and sends a sensing voltage to the controller, telling it the brakes are applied. This allows the transmission lock to disengage so you can shift out of Park. When that switch fails, the lock won't disengage. You need to replace that switch and you'll be fixed.
