Check the fuel system for flow operation and blockage / restriction. It sounds like the primer supplies enough for a couple of minutes then fails. What provides the supply after priming? Mechanical pump or electric? Check the manual.

Hard start cold, fine rest of day

I would be checking to see why there is power to the pump with the ignition off as this is a source to flat battery overnight.. May be a relay is still activated by a power source other than the ignition.

Have 76' dodge 440bb that is EXTREMELY hard to start. Can prime it cold and sometimes it will fire right up, other times it takes a few minutes. Drives fine once I get it going but if I turn it off and...

First 2 things to check are the choke adjustment and operation, and the raw fuel squirt from accelerator pump when throttle pumped. You should be able to see a good squirt of gas into throat of carburetor when you move throttle linkage to wide open quickly. Choke plate ( butterfly) should be in closed position for cold start, then open gradually as engine warms up. Have you checked that as of yet? With re-start problems after warm up, may be needle and seat assembly needs replacing to retain proper fuel level in bowl for starting. If they are dirty or worn, fuel in bowl runs back out. It sounds like a carb rebuild kit might be a good idea.

You can try this.
Turn on the key and listen for the fuel pump to prime.
It will sound like a gentle hum. when it stops, turn the key off. wait a few seconds and do it again. Do this three or four times, but don't try to start it. after the third or fourth time you turn it off, try to start it. It may not be priming the fuel injectors fast enough.
