You only need a 12v test light about 10 bucks or less. Hook test light to the door lock wires. If test light momentary lights when lock button is activated then the lock solenoid or actuator is bad. If not then wiring or fuse problem.

Most likely the door lock actuator has failed. To check you will need to pull the inner door panel, there are plenty of videos on the web that can show you how to do this. Once the panel is off find the two wires that go to the lock actuator. Use a volt meter and back probe those wires. You may need a high end meter like a Fluke or something equivalent to catch the signal when it happens. One the meter is hooked up press the lock and unlock button and see if there is voltage going to the actuator. If there is than replace it. If there is not then you will need to check wiring, most likely in the harness that runs in between the door and the body.

Like Brad said, check the ground. THAT's the first thing I always do is to be sure all grounds are clean and connected. This is a mechanics rule of thumb.

Bad grounds usually make things operate funky. If you hit the lock button and the radio turned off that would be a bad ground. If you hit the lock button and the windshield wipers started working that would be a bad ground. When something stops working all together the ground is the last suspect unless the ground wire itself is open. In that case you would find power on both wires ever time the lock or unlock was used. Since there is no path to ground the load can not be used by the device hence there is power everywhere before the open.

A combination of Jeremy and Dennis' solutions as far as troubleshooting and with what. An inexpensive test lamp probe does work well for this type of troubleshooting on the power side of the actuator, but also consider an often overlooked part of the circuit, the circuit return ground which will usually require an inexpensive analog meter to check from the actuator connector. Your local dealership should be able to give you the wire color codes at the actuatator connector for your tests as well as the physical location of the ground point from the service manual.
You did not mention if the actuator was making any sound as if trying to operate, if it makes a noise, you may just have binding linkage between the actuator movement and the drivers door lock handle, if not that, then the actuator solenoid is just worn out and in need of replacement.
Best of luck.

The lock/unlock switch will usually fail before the actuator fails.

The person sai pd the other doors lock and unlock with both the bottom and the remote. Pretty much rules out the lock/unlock switch.

2005 Chevy Equinox Driver & Passenger Door Unlock Relay. How To Remove & Install. They Are Power Locks. Front Doors Must Roll Windows Down Use Hand To Open Thanks. Ms. J. C.

Door Lock and Unlock Operation
When a door lock switch is activated in the lock or unlock position the BCM will receive a voltage signal on either the door lock switch lock or unlock signal circuits.
The BCM, upon receipt of a lock switch lock or unlock signal, will switch the appropriate lock relay control circuit(s) to battery positive voltage. This will energize the appropriate lock or unlock relay(s). The relay will supply voltage to the door lock actuator lock or unlock control circuits to one side of the lock actuator. Since the opposing side of the lock actuators are connected to ground through the other lock actuator control circuit and the normally closed contacts of the lock or unlock relay, the doors and liftgate will then lock or unlock appropriately.
Three relays are used to operate the lock. Driver door unlock, passenger door unlock and all door lock. This is done to isolate the driver door lock actuator so it can be unlocked by itself using the keyless entry transmitter.
Your door locks are controlled by the BCM - body control module (a computer ) . An a scan tool is a must to diagnose system operation . Your best bet would be to take your vehicle to a qualified repair shop . The only way you could try to fix this is by changing parts ! Suppose you find an replace a door lock/ unlock relay an that doesn't fix it ? There could be DTC'S stored in the BCM . Do you know what a DTC is ?

Test to see if you have power going to the door lock motor, if you do, then motor is bad, if you don't then there is a shorted wire or switch

I just bought an equinox 2005 LS and I dont know how to lock the rear door.

To unlock the liftgate, press the unlock button on the Remote Keyless Entry (RKE) transmitter twice or use the power door lock switch.
To lock the liftgate, press the lock button on the RKE transmitter or use the power door lock switch. The liftgate does not have a key lock cylinder.
To open the liftgate, pull up on the handle located in the center of the liftgate.
When closing the liftgate, use the molded handles to pull the liftgate down. Push the liftgate closed until it latches.

Power door locks and driver side power window not working. Keyless remote will not open doors although when pressed, the interior lights come on so I don't believe it is an issue with the fob battery....

You can do it yourself.

Couple of times when you replace the remote battery, you might have them not working/ responding but just follow the procedure below.

Stand close to the car, hold down the open and close knobs for about 10 seconds. Press them at the same time for 3 seconds; then press the open button as the car opens.

The above procedure is applicable to a lot of car brands. It does not matter how many knobs you might have on the remote fob.

I have carried this procedure out on a lot of brands ranging from Honda, Toyota, Rover, Ford, etc. and it works perfectly well.

Good luck and Rate this.

Cheers
