Question about 2000 Toyota Sienna
All other buttons will cause the right side sliding door to open. Will not open when the door handle is pulled.
Check the inside of the door for the position of the child door lock. it might be in the wrong position.

Hi my name is Peter, Is there any resistance on the door handle or does it move more freely than before? A lot of the time when something like this happens the door handle or a piece of he linkage inside the door has broken or come loose for the mechanical portion but the electrical is still functional. If the handle moves freely you may want to take the door panel off and get a better look but be careful where you put your fingers because if the electrical can still get voltage back there you may accidently activate it with you disconnect the positive side of the battery to be safe might be good idea. Depending on how popular the vehicle is and the age of it you may be able to get a replacement handle if you need one at a local parts store. If not then of course u could check online or hit the dealers parts counter.. I just replace a handle on my sons extended cab truck because the handle broke inside but I was lucky and only had two bolts to take loose lifted it out of the frame and installed the new one from Advanced auto in about 10-15 minutes. Good luck let me know what you find.

Sounds like something isn't right.Try to disconnect the battery for a few minutes and see if that helps. Some times the ecu gets haywired and disconnecting the battery might reset it.And if it is the original battery you might want to get it tested.

SOURCE: Rear Door Handle Broken
I had the same problem, and here is the solution:
It includes step-by-step instructions and photos. I printed it and headed to the garage. I while later (hours actually) it was fixed. I did end up going to the hardware store and buying the EXACT size screw they recommend. I went with a screw rather than a nut and bolt. Worked fine.

I replaced both of my sliding door handles this weekend. It was actually pretty easy. Cheapest I could find the handles was a little over $70 each. The interior panel is held on with one screw and 10 nylon clips. The one screw is in the cup holder. Put you hand in the cup holder and push out at the bottom. It pops right off exposing the one screw. I started on the clips from the top at a corner. Seemed to be the easiest place to slide a screw driver under. Once I got the screw driver in, I started working my fingers under the panel. The clips run mostly along the top and bottom about 6 inches from the edge. I made an effort to apply the force as close to the clips as I could to reduce flexing the panel. Once I got the first clip to pop out the other followed pretty easy and the panel lifted right off. Carefully peel back the plastic barrier. The exterior handle is held on with two 10mm bolts. Connecting the short cable was pretty obvious once you could see everything. Now I have to teach everyone to open the side doors very gently! Hope this helps.

Have 2005 Sienna and its Driver side door was not openning. Cheap dealer fixed it himself in front of me. After opening the doors plastic assembly while sitting inside with a screw driver. Then he bent the clip towards left where the metal wire is connected. If you guys want to see what he did i would record a video and upload for you guys. Let me know at firstname.lastname@example.org
