Question about 2000 Dodge Durango
Loosening the black plastic fender skirt by removing a couple of screws and pulling it out of the way shouldgive you access to the screw. Rmoving it probably won't be too much of a chore.

On the upper grill side of the headlight assy. there
should be 1 or 2 push pins that hold the seal on the
headlight assy to the front of the fender. Also check
for a guide/push pin on the inner top of the assy on
the outer fender side. That should be all there is to
hold the headlight in place. Hope this helps.

Not sure what your working on but to replace a headlight assembly, you shouldn't have to remove fender. Please give more details of vehicle for confirmation.
