A simple CONTINUITY test on electrical parts is a sure way of finding a junk part without spending hundreds of dollars on parts you didnt need in the first place, Dont use a mechanic who has given you a reason NOT to use him ,Ie '' has costed you senseless amounts of money and NOT a sure fix on your car !! .

Check all the above. The most common issue will be a failed injector.

It is possible. inspect the entire ignition system as well for faults. There could also be a leak in the intake manifold gasket.

back pressure. check the intake for damage and leaks. this has nothing to do with the misfire in the cylinder.

    the vacuum lines that is..

when a misfire occurs in a single cylinder, it normally is a injector or ignition issue.

Is the check engine light on?

Could have a bad pump. that wouldn't cause a misfire in the cylinder. those vacuum hoses could just be worn.

Check obvious first, starting at plug wire, to make sure it's getting spark to plug, then pull plug to see how/if, it appears to be firing. No spark to wire, check coil. If spark is good to plug, and plug firing, time to check if fuel getting there, and if no, could be caused by faulty oxygen sensor, bad injector, just as starters.
