-   I am having the same problem, I drive 100 miles a day have a 2000 es 300 with 130K on it and it wont shift into overdrive after driving it for 45 minutes. The local transmission guy said I needed a new transmission and quoted me $3500-4000--does this sound right???? That is alot of money to put into a car that is 9-10 years old.
I bought the car 11 months ago because they were supposed to be great cars--I have never had to replace a transmission-   funsister

Lexus's have a complicated system of electronics that control the function of the transmission.Under normal operating conditions the transmission is controlled by the ecu and by a secondary system called a drivetrain control module. usually when a fault occurs it sends a code to the ecu and the check engine light comes on.
If this light does not come on then typically the electronic modulator is at fault and needs to be replaced.
If the check engine light comes on when the vehicle is hot i need this code to guide u further for the repair.
If u want to attempt the repair , be wary. its messy and takes time to do to do it right.

Usually top end gears go out first. Sounds like you transmission is in premature stages of failure. I would get an internal inspection of your transmission. Problems like this tend to get more expensive if you dont catch the early.
