I had a similar problem with a sienna and sometimes the cable ( inside the door) between the handle and the latch gets stuck in the sleeve. Kind of like the brake cable on a bike.
take the panel off by removing all the screws you can see on the panel and pull the panel off. There will be plastic pins holding it. Some of them may pop off and some will break off.
Drip oil down the sleeve so the cable slides freely. I used bike chain oil. Do not use WD40. This is **** and will not work for very long. Try the handles a few times before you put the panel back.

if your asking whether or not it is a pressed in bearing or a bolt in bearing, it is a pressed in bearing. The knuckle will need to be removed and brought to a shop to have the bearing pressed in and out. It is not unusual for the hub (the part that has the wheel studs in it) to be bad as well.

U1001-This Relates to CAN communication line - signal malfunction
This code usually is in regards to the code reader unable to communicate correctly.
Is the vehicle operating correctly?Is the engine check or any other light up?If not ignore,just get the garage to clear the code if it will.
I hope this helps.if needed get back to me with findings,and I will do some checks myself on this to be 100%

that is a code for the A/F sensor. that is a air/fuel ratio sensor aka front 02 sensor. used to be called front 02 sensor, but nowadays they call them A/F sensor.
