Could be a bad plug. switch plug 1 with 2 if it follows its the plug.
could be a stuck injector, use a long tool like a screw driver and hold it up against your ear should hear a clicking repetively compare to the injector next to it. should sound the same. if not tap on injector 1 with a punch and small hammer. If it is stuck it works 90 % of the time. But there are many other things that could be wrong

Your engine sits in the engine compartment sideways. So if you are standing on the passenger side of the engine compartment looking across to the drivers side....number 1 cylinder will be closest to you on your left followed by 3 and 5. Number 2 will be the closest to you on your right followed by 4 and 6. So if you are standing in front of your car the 3 closest to the radiator from left to right are 2,4,6, and the 3 at the back of the engine from left to right are 1,3,5.

Miss firing and /check engine light fregrently comes on

These have a lot of problems with ignition coils, so replace the spark plugs with the proper Motorcraft Spark plug, and a scanner will be needed to show which cylinders are misfiring, replace those coils. Make sure the misfiring cylinders when you take the coils out , that there isnt any water/coolant in the spark plug well.

Miss fire on #3cyl. where is #3 located

Number 3 is the 3rd plug back on the passenger side. Coils are notorious for going bad, I would recommend replacing the spark plugs, while you are there. Use Auto Lite, or Motorcraft Platinum Spark plugs only.

Spark plug wires crossed on 4.3 vortec

Not sure if this is a question or a comment. The firing order will be cast on the front half of the intake manifold .1-6-5-4-3-2. Looking from the fron end the closest two terminals on your cap,the one on the right is #1 the going clockwise #6 is the other on the left and so on.The drivers side of the motor in 1-3-5 and the passinger is 2-4-6.Well hope this helps and good luck.

I have a miss fire

Recheck the firing order; 1-6-5-4-3-2

On the distributor passenger side radiator to fire wall. 2-4-6-coil
On the distributor drivers side radiator to fire wall. 3-1-5

Cylinder passenger side radiator to fire wall. 2-4-6
Cylinder drivers side radiator to fire wall. 1-3-5

Good luck and also check the distributor cap again for proper seating.
