I had that problem on the rx until today. I went to a Toyota mechanic he said turn the car on I did he looked at the over drive light it was on and said overdrive off. He had me push the over drive button on the shifter ans said drive the the rx300 it worked not quite 2500 RPM at 65mph I was happy. Maybe your overdrive off light on the speedometer console is burnt out.

Doug (the Frog) Thomas I am very happy now.....

i just had that problem recently, your car is running on safe mode, most likely your knock sensor is bad, so your car is running very rich and the gears not shifting. on the lexus rx300 their are 2 knock sensors on top of the block and under the intake manifold. let me know if you need help on how to replace it. i'll guide you through. email at part is about 180 a piece. and you have two. diagnose it with obd2 diagnostics and it will show you code on which one to replace if you dont have much money but its better to replace both at the same time.

When i shift car from P to R or D the rpm suddenly goes to zero and then suddenly goes to approx 2000 rpm.

that should not effect the driving of the car,it's do to the loss of connection in the gear shifter.

Engine sounds like it is screaming RPM's and sounds like it needs to shift down but stays reved up

Can you count the number of shifts ?
2nd speed at 10 - 3rd speed at 20 - lockup and overdrive at 40 ?
Normal RPM for most cars is around 2000 at 60mph.

With normal acceleration the 1st to second shift point should be around 2500-3000 rpm. Most of your shifts under moderate acceleration will take place at this same point or when there a light load on the transmission. Under hard acceleration the RPMs will climp higher due to increased power needs and the computer will shift when less power is needed or the RPMs climb to an unsafe speed say5000-6000rpms (depending on where yours redlines). Also going up a steep hill will raise the RPMs higher and goind down a hill will cause a faster shift at lower RPMs. These can always be altered by an individual owner by adjusting shift points, using performance parts and if you have problems (low tranny fluid/trans issues). Your trans RPMS are fine and will vary depening on how your foot accelerates. The shift pattern should be the same every time you drive slow/medium/fast/, look for changes in those patterns you will also catch a problem early.

Transmission shifting problem

seems you have a transmission failure problem. You should repair your tranny as soon as possible. As it only will got worse and cost more in the future if you choose to ignore it now.
